Nat Cancer Scientists discover optimal anti-cancer T cell subset

On May 22, 2024, a study published in the international journal Nature Cancer is entitled "Identification of a clinically efficacious CAR T-cell subset in diffuse large B cell lymphoma "by dynamic multidimensional single-cell profiling, scientists from the University of Houston and other institutions have identified a new class of T cell subsets that may improve the prognosis of patients receiving T cell therapy.

CAR-based on T cells, T cells immunotherapy in shown tremendous value to resist and eliminate cancer, and this strategy can also activate the body's immune system in patients with engineering modification of the patient's own T cells to recognize and attack and kill cancer cells, so that the body's own T cells can become live. Although T-cell therapies have revolutionized the treatment of human cancers, not all patients have a response to these therapies. Therefore, a better understanding of the properties of engineered T cells is necessary to improve clinical responses.

In this study, the researchers used a TIMING (Timelapse nano trap grid time-lapse Imaging Microscopy Imaging Microscopy in Nanowell Grids) method, it can be applied to evaluate visual AI cell behavior, movement and ability. Research results show that the CD8 conformal T cells can migrate and continuous destruction, and have a balance of mitochondria and lysosome volume. Using independent data sets, the researchers verified the CD8 conformal T cells: (1) there before manufacturing, and to use axicabtagene ciloleucel associated with individual clinical response, (2) after treatment with CAR T cells in the individual longitudinal persist, (3) can be carried out within the solid tumor amplification migration of tumor cells to dissolve cells within the tumor. In conclusion, in this paper, the results of the study clarified the cell function evaluation of multimodal integration in the discovery and application of CD8 fit T cells for cell therapy has the best adaptability of T cell subsets in the aspects of value and power.
